How often do we consider membrane fluidity when optimizing liposome or LNP manufacturing?
Is it time to rethink lipid-based drug delivery through the lens of membrane biophysics?
This mini-review highlights the essential role of membrane fluidity in lipid-based drug delivery systems such as liposomes and lipid nanoparticles. Fluidity, regulated by temperature, lipid composition, cholesterol content, and surface tension, directly influences drug loading, particle size, stability, and targeting. The authors emphasize that recognizing and controlling membrane fluidity as a Critical Quality Attribute (CQA) can significantly enhance formulation development and manufacturing robustness, improving product performance and process consistency.
